I need to design a website for making it responsive and mobile friendly. The current layout is table based. will it be easy to make it mobile friendly just by tweaking CSS (media queries) or need to change the HTML structure to table-less first?

Toss it and start over
There is nothing socially redeeming about that markup. Nor is there a sane method of making the site small-device friendly.
The need for table layouts died with the development of css2 in circa 1997 or 98 and html4 in 1999. So you're trying to hitch this century's technology wagon to last century's lame horses.
